A copper(II) complex containing the ceftobiprole drug and 1,10-phenanthroline (phen) has been synthesized and characterized by UV–vis, FT-IR and mass spectra, and elemental analysis. The binding interaction between [Cu(cef)(phen)Cl2] complex and human serum albumin (HSA) was investigated using absorption, fluorescence emission and circular dichroism spectroscopies, and molecular docking. Thermodynamic parameters (ΔH < 0 and ΔS < 0) indicated that the hydrogen bond and van der Waals interactions played main roles in the binding of complex [Cu(cef)(phen)Cl2] to HSA. The results of CD and UV–vis spectroscopy showed that the binding of [Cu(cef)(phen)Cl2] to HSA induces some conformational changes in HSA. Displacement experiments predicted that the binding of [Cu(cef)(phen)Cl2] complex to HSA is located within domain III, Sudlow’s site 2, and these observations were substantiated by molecular docking studies. 相似文献

At high Ph(3)SnH concentrations, the O-directed hydrostannation pathway dominates, and 2 is formed with good selectivity (ca. 11.1:1). This paper describes the instrumentation developed by Pierre and Jacques Curie in the 1880s to perform very precise charge and low current measurements. Using this set-up, which is nearly as precise as most present digital electronic charge measurement apparatuses, Jacques Curie established the absorption current law for insulating materials. A detailed analysis is made of the experimental path followed by Marie Curie from the choice of her research subject to her first discoveries, using the same Curie quartz–electrometer set-up. From the Curie “discovery notebooks” and a modern reconstitution of this experiment, it is possible to reconstruct what were her main experimental difficulties concerning the measurements of the picoampere leakage currents produced by the radioactive rays. We will underline that the availability for Marie Curie of a precise ready-to-use electrostatic measurement technique played an important, if not decisive, part in her career and her research. A more general issue addressed here is that laboratory techniques are not only a prerequisite of modern scientific work, but in a great part influence knowledge production. 相似文献

The aim of this work was to optimize the conditions for in vitro synthesis of (1→3)-β-D-glucan (callose) and cellulose, using detergent extracts of membranes from hybrid aspen (Populus tremula × tremuloides) cells grown as suspension cultures. Callose was the only product synthesized when CHAPS extracts were used as a source of enzyme. The optimal reaction mixture for callose synthesis contained 100 mM Mops buffer pH 7.0, 1 mM UDP-glucose, 8 mM Ca2+, and 20 mM cellobiose. The use of digitonin to extract the membrane-bound proteins was required for cellulose synthesis. Yields as high as 50% of the total in vitro products were obtained when cells were harvested in the stationary phase of the growth curve, callose being the other product. The optimal mixture for cellulose synthesis consisted of 100 mM Mops buffer pH 7.0, 1 mM UDP-glucose, 1 mM Ca2+, 8 mM Mg2+, and 20 mM cellobiose. The in vitroβ-glucans were identified by hydrolysis of radioactive products, using specific enzymes. 13C-Nuclear magnetic resonance spectroscopy and transmission electron microscopy were also used for callose characterization. The (1→3)-β-D-glucan systematically had a microfibrillar morphology, but the size and organization of the microfibrils were affected by the nature of the detergent used for enzyme extraction. The discussion of the results is included in a short review of the field that also compares the data obtained with those available in the literature. The results presented show that the hybrid aspen is a promising model for in vitro studies on callose and cellulose synthesis. 相似文献

Accumulation of protoporphyrin IX (PpIX) was investigated in normal skin and UV-induced tumours in hairless mice after topical application of a cream containing 2, 8 or 16% of 5-aminolevulinic acid methyl ester (ALA-Me). Higher levels of PpIX were measured in tumours compared to normal skin. The maximal amount of PpIX was reached at 1.5, 3 and 4 h after 2, 8 and 16% ALA-Me application, respectively. Higher tumour to normal skin PpIX fluorescence ratios were measured after application of 8 and 16% ALA-Me than after application of 2%. After irradiation with a broad spectrum of visible light from a slide projector, more than 90% of PpIX was bleached by fluences of 36 and 48 J/cm2, at fluence rates of 10 and 40 mW/cm2 respectively. At these fluences, the PpIX photobleaching rate was significantly higher (P<0.05) in normal mouse skin than in tumours. In addition, for a given fluence, more PpIX was photobleached at the lower fluence rate (10 mW/cm2) than at the higher fluence rate (40 mW/cm2) in normal skin (P<0.001) as well as in tumours (P<0.05) after exposure to 24 J/cm2 of light. In conclusion, the highest tumour to normal skin PpIX ratio was observed 3 h after application of 8% ALA-Me, suggesting that light exposure should be performed at this time in order to achieve an optimal PDT effect in this tumour model. 相似文献

Background
Annexin A7 is a member of the annexin protein family, which is characterized by its ability to interact with phospholipids in the presence of Ca2+-ions and which is thought to function in Ca2+-homeostasis. Results from mutant mice showed altered Ca2+-wave propagation in astrocytes. As the appearance and distribution of Annexin A7 during brain development has not been investigated so far, we focused on the distribution of Annexin A7 protein during mouse embryogenesis in the developing central nervous system and in the adult mouse brain. 相似文献30.
